Will they work with Google Maps or Mapbox?

Yes, custom markers in both accept PNG images. Your developer sets the size and anchor point.

Can I get a selected state for each pin?

Yes, a selected or active version counts as a separate icon in the package.

Can you add cluster markers?

Yes, a cluster marker can be one of the icons. Numbers on it are added by your map code.
